摘要:
目的 探讨激活葡萄糖脑苷脂酶(GBA)对帕金森病患者皮肤成纤维细胞自噬活性的影响。方法 取32例帕金森病患者的皮肤标本进行成纤维细胞体外培养,运用含有雷帕霉素及不含雷帕霉素的GBA培养基分别进行体外培养,用蛋白印迹法检测自噬体的经典标记物微管相关蛋白的表达。结果 雷帕霉素处理后 Beclin 1和LC3-Ⅱ/LC3-I蛋白的表达水平增高(P<0.05),而a-synuclein表达水平则下降(P<0.05)。结论 激活GBA可以强化自噬水平,降低a-synuclein蛋白的表达。
Abstract:
ObjectiveTo investigate the effect of activation of glucocerebrosidase on autophagotrophic activity of skin fibroblasts in Parkinson’s patients.Methods 32 cases of Parkinson’s disease were selected. Fibroblast cells were cultured in vitro with rapamycin or without rapamycin GBA culture medium respectively. Microtubule associated protein phage classic markers expression were detected by protein imprintting method.Results Beclin 1 and LC3 - Ⅱ/LC3 - I protein expression level increased(P<0.05), while a - synuclein expression level decreased(P<0.05).Conclusion GBA activity can strengthen autophagy and reduce a - synuclein protein expression.
