摘要:
目的 探讨长链非编码RNA MEG3,HOTAIR和MALAT-1对垂体腺瘤发生发展和侵袭的作用。方法 应用实时定量逆转录聚合酶链式反应(qRT-PCR)检测MEG3,HOTAIR,MALAT-1在正常脑组织、非侵袭性和侵袭性垂体腺瘤中的表达情况,并分析3组之间的表达水平差异。结果 MEG3表达水平在正常脑组织、非侵袭性垂体腺瘤和侵袭性垂体腺瘤中逐渐降低,而HOTAIR表达水平逐渐升高(P<0.05)。MALAT-1的表达水平在3组样本之间没有统计学差异(P>0.05)。MEG3的表达水平与垂体腺瘤侵袭性生物学行为有相关(P<0.05),HOTAIR的表达水平与侵袭性生物学行为和肿瘤大小有关(P<0.05),而MALAT-1的表达水平与年龄、性别、分类、生物学行为和肿瘤大小之间没有显著关系(P>0.05)。结论 MEG3和HOTAIR的表达水平异常可能与垂体腺瘤的发展和侵袭密切有关。
Abstract:
ObjectiveTo study the roles of long non-coding RNA MEG3,HOTAIR and MALAT-1 in the development and invasion of pituitary adenomas.Methods Real-time quantitative reverse transcription polymerase chain reaction(qRT-PCR)was used to detect the expression levels of MEG3,HOTAIR and MALAT-1 in normal brain tissues,non-invasive and invasive pituitary adenomas,and the expression level differences were analyzed among them.Results The expression level of MEG3 was gradually decreased from normal brain tissues to non-invasive pituitary adenomas,from non-invasive pituitary adenomas to invasive pituitary adenomas,while HOTAIR lncRNA levels were gradually increased with statistically significant difference(P<0.05).The expression level of MALAT-1 was not statistically different among the three groups(P>0.05).There was a significant association between expression level of MEG3and biological behavior of tumor invasion(P<0.05).The expression level of HOTAIR was correlated with invasive biological behavior and tumor size(P<0.05),but there was no significant correlation between the expression level of MALAT-1 and age,sex,classification,biological behavior and tumor size(P>0.05).Conclusion The abnormal expression levels of MEG3 and HOTAIR might be closely related to the development and invasion of pituitary adenomas.
