摘要:
目的 探讨颞叶癫痫(Temporal lobe epilepsy,TLE)患者外周血单核细胞(Peripheral blood mononuclear cells,PBMCs)上的谷氨酸受体(Ionotropic glutamate receptors,iGluRs)表达水平变化。 方法〖KG1〗 纳入2019年1月-2022年6月在本院接受治疗的17例TLE患者和21名健康对照者作为研究对象;从所有研究患者及健康对照组中收集3 mL晨起静脉血样品,并通过密度梯度离心法分离PBMCs;通过聚合酶链反应(Polymerase chain reaction,PCR)检测PBMCs中iGluR基因表达水平;蛋白质印迹分析PBMCs中AMPA离子能谷氨酸受体3(AMPA receptor 3,GluA3)蛋白表达水平;使用荧光测定法来测量外源性腺嘌呤核苷三磷酸(Adenosine triphosphate,ATP)对单核细胞内Ca2+稳态的影响。 结果 与健康对照组比较,TLE组在焦虑和抑郁量表上表现出更高的分数(P<0.05),并且PBMCs中α-氨基-3-羟基-5-甲基-4-异恶唑丙酸(α-Amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid,AMPA)受体亚单位GluA3 mRNA表达水平显著降低(P<0.001);TLE组PBMCs中GluA3蛋白表达水平显著低于3健康对照组(P<0.001)。与健康对照组比较,TLE组的PBMCs在第2个周期和第3个周期显示Ca2+水平显著降低(P<0.05)并且在全部周期显示峰值振幅显著降低(P<0.05)。 结论 TLE患者PBMCs中特定iGluRs亚单位GluA3受体的表达水平降低,并可能影响单核细胞的钙稳态。
Abstract:
Objective〖WTBZ〗 To investigate the expression of glutamate receptors (iGluRs) on peripheral blood mononuclear cells (PBMCs) in patients with temporal lobe epilepsy (TLE). 〖WTHZ〗Methods 〖WTBZ〗 In this study, 17 TLE patients and 21 control groups who were treated in our hospital from January 2019 to June 2022 were included as study subjects. 3 ml venous blood samples were collected from patients and the control group, and PBMCs were separated by density gradient centrifugation. The expression of the iGluR gene in PBMCs was detected by PCR. The expression of GluA3 protein in PBMCs was analyzed by Western blot. Fluorescence assay was used to measure the effect of exogenous ATP on intracellular Ca2+ homeostasis of monocytes. 〖WTHZ〗Results 〖WTBZ〗 Compared with the control group, TLE patients showed higher scores on anxiety and depression scale (P<0.05), and the expression of AMPA receptor subunit GluA3 mRNA in PBMCs decreased significantly (P<0.001). The expression of GluA3 protein in the PBMCs in TLE group was significantly lower than that in the control group (P<0.001). Compared with the control group, PBMCs of TLE patients showed a significant decrease in Ca2+level in the second and third cycles (P<0.05), and a significant decrease in peak amplitude in all cycles (P<0.05). 〖WTHZ〗Conclusion 〖WTBZ〗 The expression of GluA3 receptor on of specific iGluRs subunits in PBMCs in the TLE patients decreases, which may affect the calcium homeostasis of monocytes.
